Cysteine cathepsins are proteases belonging to the papain family. Predominantly located in lysosomes, their primary physiological roles include the degradation, inhibition, and processing of other proteins. The human genome encodes 11 cysteine cathepsins: B, C, F, L, K, V, S, X/Z, H, W, and O. They all share a conserved fold with catalytic residue pairs Cys25 and His159 (according to papain numbering) in their active site. GAG binding can alter their enzymatic stability, localization, and activity, making these interactions a critical target for biochemical modeling.
